Abstract

An alternative for the in vitro propagation of plants is the use of Temporary Immersion Systems; however, limited information is available on its use in cultivation of yam, and there are no references to the Chinese White clone. The objective of the article was to determine the content of mineral nutrients and metabolites present in the multiplication culture medium of Dioscorea alata clone Chino Blanco in Temporary Immersion Systems. To determine the content of total nitrogen, calcium and magnesium, samples of the culture medium were taken at the beginning and at the end of the six weeks of culture. The total nitrogen content will be reduced by the colorimetric method with the Nessler reagent, while calcium and magnesium will be reduced by the volumetric method with Ethylene-Diamine-Tetracetic. To corroborate the presence of groups of metabolites, negative and positive controls were used with culture medium after 28 days of culture. Different tests were used for the identification of carbohydrates, alkaloids, phenols, quinones, catechins, free amino acids, flavonoids, triterpens and steroids. In the culture medium, the total nitrogen content was exhausted, while the concentration of calcium and magnesium increased; In addition, the presence of some metabolites exuded to the culture medium such as carbohydrates, catechins, quinones and phenols was identified.
