Abstract

The culture of sweet potato (Ipomoea batatas (L.) Lam.) constitutes a important food worldwide, being necessary the use of biotechnological techniques to facilitate a greater availability of planting material with the increase the use of in vitro micropropagation techniques, specifically, the somatic embryogenesis, would help to establish quality seed banks. In the present investigation, explants of leaf blades of tuberous root shoots belonging to clones CEMSA 78-354, INIVIT B 90-1, INIVIT B 93-1, Yabú-8 and Jewel were used, disinfected with sodium hypochlorite (1%,) for 15 minutes, subsequently, the potentially embryogenic callus were formed, the somatic embryos were obtained, they matured, germinated and converted into seedlings. Being used the culture medium proposed by Murashige and Skoog (1962) and for the formation of potentially embryogenic callus, 2,4-D (0,50 mg.L-1) and 6-BAP (0,25 mg.L-1) and the formation of somatic embryos was achieved with the addition of 2,4-D (0,2 mg.L-1), with no significant differences between the clones in both experiments. Maturation of somatic embryos was achieved with the addition of abscisic acid (ABA) a 1,0 mg.L-1 and with thidiazuron (TDZ) a 0,25 mg.L-1 the highest results in germination; meanwhile, the conversion into seedlings was obtained with the use of giberellic acid (GA3) a 10,0 mg. L-1.
