Effect of explant type and disinfection in the establishment in vitro of Mulberry criolla variety (Original)

The mulberry crop constitutes an alternative source of high nutritional content for the animal feeding under the tropical conditions. In this species diverse propagation protocols have been developed, however, the presence of endogenous contaminant in the explant affects the in vitro establishment efficiency. The investigation was carried out with objective to define the explant type for use in the in vitro establishment phase and to optimize a disinfection method of Criolla variety. They took apical bud as well as nodal segments of basal and central from donating bud explant. Then sodium hypochlorite (1.0 %) for 1 min. alone or combined with ethanol 70 % were evaluated. As result the apical bud demonstrated the less contaminant (20.6 %) that favored the efficiency (60.3 %) during in vitro establishment. After ethanol 70 % for 1 min., followed 10 min at sodium hypochlorite 1.0 %, it was possible to decrease the contamination up to 17.0 % and increased the explant survival (71.0 %) of mulberry Criolla variety.
